Genflow nears key data readout for SIRT6 gene therapy platform

Genflow Biosciences is approaching an important stage in the development of its SIRT6 gene therapy platform, with further data from its aged-dog studies expected shortly and a new delivery partnership now in place.

The company said early effects seen in its Sarcopenia and Longevity in Aged Beagles study were still present three months after treatment. The blinded trial includes 24 beagles aged over 10 years and is testing two naked DNA doses, an AAV8 viral vector treatment and an untreated control group.

Initial results released earlier in 2026 showed that all three treated groups had better survival during the dosing period than the control group. No adverse events were reported. Genflow later confirmed that the observed improvements remained visible three months after dosing and that no new safety concerns had emerged.

The findings are still preliminary. The company is continuing to analyse muscle biopsies and methylation clock data, which should provide a clearer view of whether the treatment is affecting biological ageing, muscle condition or both.

Genflow’s lead therapy, GF-1002, uses a variant of the SIRT6 gene linked to exceptional human longevity. SIRT6 is involved in DNA repair, genomic stability and the cellular response to stress. The company is developing a centenarian-associated form of the gene rather than the standard version.

The current study is also testing different ways to deliver the therapy. Naked DNA is simpler and may be cheaper to produce, but uptake into cells can be limited. AAV8 viral vectors can support longer-lasting gene expression, although repeat dosing and existing immunity may create constraints.

Genflow is also developing a messenger RNA approach using lipid nanoparticles. This method would produce temporary gene expression and may allow repeat administration. Its value will depend on whether the formulation can reach the required tissues efficiently.

To advance this work, Genflow has entered a collaboration with Acuitas Therapeutics. Acuitas will formulate Genflow’s SIRT6 messenger RNA using its lipid nanoparticle technology and return the finished formulations for preclinical testing.

Acuitas is funding the work, making the programme non-dilutive for Genflow shareholders. The deal is not expected to generate immediate revenue, but it gives Genflow access to specialist delivery technology without requiring it to carry the full development cost.
