Genflow Biosciences has strengthened the development case for its SIRT6 gene therapy platform after its Sarcopenia and Longevity in Aged Beagles clinical trial met its primary endpoint, providing new evidence that the company’s technology can influence biological markers associated with ageing.
The SLAB study evaluated Genflow’s centenarian SIRT6 therapy in beagles aged over 10. Treated animals recorded a reduction in biological age compared with untreated controls, measured using the GRIM DNA methylation clock.
The result gives Genflow an additional proof point for a development strategy centred on SIRT6, a gene involved in cellular processes associated with ageing. The company is focused on a specific variant found more frequently in centenarians than in the wider population, with its lead compound GF-1002 designed to deliver this variant directly.
The trial was randomised and blinded and included 24 aged beagles divided into four groups. Two groups received different doses of naked DNA treatment, another received a single dose delivered through an AAV8 viral vector, while the fourth acted as a saline control.
Meeting the primary endpoint follows earlier indications from the programme. Genflow had previously reported positive interim muscle data in February, followed by evidence of sustained safety and efficacy at a three-month follow-up in April. The company has also said treated animals improved across several secondary functional and observable measures, although the detailed results have not yet been released.
